Wheel Repair Questions

I am  attempting to replace broken / missing spokes.  I plan to wrap interior
of rims (over spoke holes) with 10 mill pipe wrap.  I also will cover spoke
ends inside grease areas with caulking.

Any hints on tightening spokes?  I plan to use the sound method... hit and
compare sound with existing spokes!

What is the best caulking to use??? 

What else should I do while these are apart???
