Not not plug that hole. It is the drain hole for the bell housing. No doubt that your rear main scroll seal is leaking a bit and finding it's way out there. If you plug the hole you will just fill the bell-housing with oil though I doubt it is "Water tight" enough to do so. Thought it might fill enough to touch the flywheel edge and make a mess, not to mention an oil soaked clutch plate.
