Are these SU's or Solexes? If solex, there could be something wrong
with that one barrel, but if SU's, a problem should affect both 3 and 4.

My bet is on the distributor!

If the shaft has a wee bit of wobble, it'll cause one or more plugs to
misfire. Pop the cap and rotor and check for play in the shaft. If
you're really ambitious (I don't really recommend this), pull the unit
and pedestal, pull the gear, turn it exactly one half turn (you may have
to turn the oil pump too, but I don't think it's offset) and drop it back.
Turn the distributor shaft exactly a half turn too, and put it all back
together. SWAP THE PLUG WIRES TOO. What you've done is moved
what plug is associated with the same physical position on the rotor.
So if it is the distributor, your problem cylinder will move!
