You can unplug the DIS while you bump the starter.  You can remove the top timing belt cover, but you'll have to disconnect the DIS anyway in order to do this.  Rotate the engine until the mark on the harmonic balancer lines up with 0 degrees, then the cams will either be lined up with their marks or exactly 180 degrees out of alignment.

I'm not sure why you are checking the timing unless you wonder if somebody put the car together wrong.  If you are concerned about "crank cancer" then that is checked slightly differently.
