The application here on the driveshaft flange to dif flange is one I've been 
using
nylock nuts on for about 25 years with never a problem on grade 5 bolts.  
HOWEVER,
the bolts and the length of the threads on them are far more important than the
nylock or nuts and lockwasshers.  There should be but no to only a few threads
inside the flange holes.  The bolt shaft is an important part of this 
connection.

Don't use any bolts with threads rolled way up the shaft so mostly threads are
located inside the flanges.
